    Hello,
    I’m fairly new to the chi.bio platform, but recently got it to run.
    I’m planning an experiment, and although my first instinct has been to use EMS as a mutagen, I am considering using the UV bulb in the reactor. That being said, all I can find about the bulb is that it’s 280nm, but nothing about intensity. I want to know before I start what a good starting point would be so that I achieve an ample level of mutagenesis. Does anybody have any experience using this and can share from their experience?

    Hi Boaz,

    I don’t know how much mutagenesis the UV LED introduces in biological terms (i.e. rate of DNA damage / LED intensity), however, if you look in the Chi.Bio paper in PLOS Biology one of the figures shows how different intensities impact E.coli in terms of slowing down their growth (which they eventually adapt to), so that may provide you with some good initial estimates of the intensities you might use.

    Hi Harrison,

    Could I just check in that figure S1N, is the UV just left on? Or is it flashed for a set time? Wasn’t completely clear. Just so I have a sensible ballpark to start testing the UV out. 🙂
