Hi there. Posting on forum as may be of interest to other users, I looked and couldn’t find this being addressed previously.
The fluorescence readout from the ChiBio is light measured / OD (i.e. fluorescence per cell). We’d actually like to measure the culture fluorescence – as a way to track the proportion of the culture that is fluorescent or not. Are the ‘raw’ Ie values stored or is there somewhere in the code we can edit to remove this /OD step?
Hello! Yes, you could just multiply the reading (e.g. FP1_Emit) by the baseband reading, and that gives you the absolute value of the reading (without normalising for excitation light/density)
